Sionna narrows its cystic-fibrosis program and cuts 46% of its workforce
Sionna will advance the SION-451 plus SION-2222 dual combination into an open-label, 28-day Phase 2a proof-of-concept trial expected to begin in the first quarter of 2027. It is not advancing SION-719 as an add-on to standard of care after that program missed its key activity endpoint.
The company also announced a roughly 46% workforce reduction and other cost-saving measures that it expects will extend cash runway into the second half of 2029. The new path is supported partly by post hoc analyses and remains unproven.
Why it matters
The reduction is an immediate negative signal for affected employees. The narrower program may concentrate remaining work in cystic-fibrosis clinical development, translational science, regulatory, and trial operations, but Sionna did not announce hiring tied to the Phase 2a plan.

PIPE-307 misses the primary endpoint in a Phase 2 depression study
Contineum said MOONLIGHT-1 did not show improvement over placebo on the MADRS total score at Day 5, its primary efficacy endpoint. The company said JNJ-5120/PIPE-307 was well tolerated with no new safety signals.
Johnson & Johnson continues to review the data and prespecified exploratory endpoints before deciding next steps. Neither company announced a staffing decision.

Oncology / FDA designation
RASONQUE gains Breakthrough Therapy status for a first-line combination
FDA granted the designation to RASONQUE with gemcitabine and nab-paclitaxel for treatment-naive metastatic pancreatic adenocarcinoma, based on preliminary Phase 1/2 data.
The combination remains investigational and is being evaluated in the global Phase 3 RASolute 303 trial. This is distinct from RASONQUE's approval in a later-line population.

Agomab reports early IPF data and maps a Phase 2 study
Agomab reported results from 10 patients with idiopathic pulmonary fibrosis treated in the Phase 1 cohort of inhaled, lung-restricted AGMB-447.
The company described generally favorable safety, low systemic exposure, high lung exposure, and ALK5 target engagement. A Phase 2 application has been submitted, but these small, early findings do not establish efficacy.

The takeaway
A strong hiring pulse does not erase program-level risk.
Sionna's restructuring and Contineum's missed endpoint show how quickly portfolio decisions can change the employment picture. RASONQUE's designation and Agomab's early respiratory data keep execution work moving, but both updates carry clear limits. Today's verified openings offer a separate view across discovery, manufacturing, quality, clinical leadership, and analytical development.
